COMET FISH


The COMET FISH program is an appropriate program to measure the DNA and FISH signals in images obtained from the single cell
gel electrophoresis (Comet assay). It requires high magnified images (objective *40, or higher) and can be applied with or without
syntactic structure analysis. It provides the measurement data of a single or of two probes (red, green labelled) and measures the
DNA distribution in the nucleus and tail as well as FISH signals located within the nucleus (DAPI stained) and outside the nucleus
if present. It can be applied
with or without syntactic structure analysis.

The measurement data of the syntactic structure analysis are described separately.

The measurement program will provide you with the following results:

Measurement data
  • mean size nucleus Vv: mean Volume fraction of nuclear area
  • percent DNA in nucleus: percent of DNA in nucleus (in relation to DNA (nucleus) + DNA (tail))
  • mean IOD in nucleus: mean integrated optical density (IOD) of DNA in the nucleus
  • neg. nuclear DNA area: nuclear area without DNA in the nucleus
  • range nuclear DNA: range of integrated optical density (IOD) in the nucleus
  • mean size tail Vv: Volume fraction of tail area
  • percent DNA in tail: percent of DNA in tail (in relation to DNA (nucleus) + DNA (tail))
  • mean IOD in tail: mean integrated optical density (IOD) of DNA in the tail
  • neg. tail DNA area: tail area without DNA
  • range tail DNA: range of integrated optical density (IOD) in the tail
  • No DNA agglutinations tail: number of DNA agglutinations in the tail
  • rel. distance tail center<> nuclear center: relative distance of center of gravity in the tail to the center of gravity in
    the nucleuas (in relation to the nuclear diameter)
  • cor. coefficient dx<>area tail DNA: correlation coefficient between area of DNA agglutinations in the tail and their
    distance to the nuclear center
  • cor. coefficient dx<>IOD tail DNA: correlation coefficient between IOD of DNA agglutinations in the tail and their distance
    to the nuclear center
  • cor. coefficient dx<>neg DNA area tail: correlation coefficient between negative area of DNA agglutinations in the tail and
    their distance to the nuclear center
  • cor. coefficient dx<>range tail DNA: correlation coefficient between IOD range of DNA agglutinations in the tail and their
    distance to the nuclear center
  • n-star tail DNA agglutinations: mean of No of neighbouring DNA agglutinations in the tail
  • rel. edge length tail DNA: mean of relative distance between DNA agglutinations in the tail
  • entropy IOD DNA agglutinations: mean structural entropy of DNA agglutinations in the tail
  • No nuclear FISH-1-: absolute No of FISH signals of the first probe (if two probes ->red, if only one probe -> red or green)
    in the nucleus
  • rel. distance FISH-1-<>nuclear center: mean of relative distance of nuclear FISH signals No 1 to the nuclear center
  • No FISH-1- tail: absolute No of FISH signals of the first probe (if two probes ->red, if only one probe -> red or green) in the tail
  • rel. distance FISH-1-tail<>nuclear center: mean of relative distance of FISH signals No 1 in the tail to the nuclear center
