Nuclei
The nuclear staining program is an appropriate program to measure antigens that are present in the nuclei. It requires moderately
to high magnified images (objective *20, *40, or higher) and can be applied with or without syntactic structure analysis. Appropriate
markers are those for proliferating (or apoptotic) cells, or sexual hormones, (Ki-67, bcl_2, oestrogen, progesterone receptors, etc.).
The measurement program will provide you with the following results:

Measurement data
- No nuclei: Number of measured nuclei
- No positive nuclei: Number of measured nuclei with positive staining
- Vv nuclei neg: Volume fraction of negative nuclei 100 *[S{(pixnu)/(pixtot)}]
- Vv positive stain: Volume fraction of all stained pixels (independent from nuclear, membrane, etc. stain)*100
- % Vv pos nuclei: percent of Vv of positive stained nuclei (Vv of positive stained nuclei/Vv all nuclei)*100
- % weak stain: Percent of weakly stained nuclei
- % mod. stain: percent of moderately stained nuclei
- % inten stain: percent of intensively stained nuclei
- mean nuclear distance: mean distance between gravity centers of nuclei
- mean nuclear area: mean area of all detected nuclei (either in pixels or in absolute square microns)
- stnd dev nuclear area: standard deviation of mean nuclear area
- mean max diameter: mean of maximum nuclear diameter
- stnd dev max diameter: standard deviation of mean of maximum nuclear diameter
- mean min diameter: mean of minimum nuclear diameter
- stnd dev min diameter: standard deviation of mean of minimum nuclear diameter
- percent pos nuclei: percent of positively stained nuclei (>10% of nuclear area)
- percent mean pos nucl area: {mean of positive-stained nuclear area/mean of total area of positive stained nuclei}*100
- mean pos nucl area: mean of area of positive stained nuclei
- stnd pos nucl area: standard deviation of mean of area of positive stained nuclei
- mean neg nucl area: mean of area of negative stained nuclei
- stnd neg nucl area: standard deviation of mean of area of negative stained nuclei
- p diff nucl area: p-value (t-test) on difference between areas of positive – negative stained nuclei;
- mean pos min diameter: mean of minimum diameter of positive stained nuclei
- mean pos max diameter: mean of maximum diameter of positive stained nuclei
- stnd pos max diameter: standard deviation of mean of maximum diameter of positive stained nuclei
- mean neg max diameter: mean of maximum diameter of negative stained nuclei;
- stnd neg max diameter: standard deviation of mean of maximum diameter of negative stained nuclei
- p diff max diameter: p-value (t-test) on difference between maximum diameters of positive – negative stained nuclei
- mean pos min diameter: mean of minimum diameter of positive stained nuclei
- stnd neg max diameter: standard deviation of mean of minimum diameter of positive stained nuclei
- mean neg min diameter: mean of minimum diameter of negative stained nuclei
- stnd neg max diameter: standard deviation of mean of minimum diameter of negative stained nuclei
- p diff min diameter: p-value (t-test) on difference between minimum diameters of positive – negative stained nuclei
- grading (%Vv * %int nuclei): quantitative grading for immunohistochemistry reaction
(percent of positive stained nuclei Np) * (positive area fraction within the nuclear area of positive stained nuclei).
